噬菌蛭弧菌噬菌斑的鉴别与纯化的研究

本文报告了使用１／５００营养肉汤双层琼脂培养基,在含有宿主菌及寄生菌混合物的软琼脂层中,蛭弧菌、变形虫及鞭毛虫等均可形成噬菌斑,但其大小、形态、扩展速度不同。鉴别方法除依据噬菌斑形态特征外,主要靠相差显微镜下的形态学观察。蛭弧菌的纯化用单斑传代,一个直径为１ｍｍ的噬菌斑含有约１０^５－６ｐｆｕ／ｍｌ（每毫升噬菌斑形成单位）。     

一株能表达987P和F_(41)两种粘附素的猪源性大肠杆菌的研究

通过直接凝集试验、免疫荧光试验、SDS-PAGE和Western印迹,对一株猪源性大肠杆菌的粘附素进行了研究.结果表明,该菌株是一株同时表达987P和F_(41)两种粘附素抗原的猪源性大肠杆菌.     

Requirement of human immunodeficiency virus type 1 nef for in vivo replication and pathogenicity.

The role of human immunodeficiency virus type 1 (HIV-1) accessory genes in pathogenesis has remained unclear because of the lack of a suitable in vivo model. The most controversial of these genes is nef. We investigated the requirement for Nef for in vivo replication and pathogenicity of two isolates of HIV-1 (HIV-1JR-CSF and HIV-1NL4-3) in human fetal thymus and liver implants in severe combined immunodeficient mice. HIV-1JR-CSF and HIV-1NL4-3 differ in their in vitro phenotypes in that HIV-1JR-CSF does not induce syncytia and is relatively noncytopathic, while HIV-1NL4-3 is highly cytopathic and readily induces syncytia. The nef mutants of both isolates grew with kinetics similar to those of parental virus strains in stimulated peripheral blood lymphocytes but demonstrated attenuated growth properties in vivo. HIV-1NL4-3 induced severe depletion of human thymocytes within 6 weeks of infection, whereas its nef mutant did not. Thus, HIV-1 Nef is required for efficient in vivo viral replication and pathogenicity.     

Purification and characterization of recombinant human pro-urokinase produced in silkworm using a baculovirus vector

Summary A recombinant baculovirus (BmNPV-pk2) was constructed by inserting the human pro-urokinase(pro-UK) cDNA into the genome of baculovirus Bombyx mori nuclear polyhedrosis virus (BmNPV) adjacent to the strong polyhedrin promoter. The recombinant virus replicated in silkworm larvae, which synthesized 30g pro-UK/ml in the haemolymph within 4 days post-infection. Purification to near homogeneity was accomplished by fractional precipitation with ammonium sulphate and immunoaffinity chromatography with an overall yield of 23% and a specific activity of 100,000IU/mg in fibrin plate assay. This purified product was comprised of a single chain protein with approximately Mr. 50kDa as determined by SDS-PAGE gel. The N-Terminal amino acids sequence revealed that the secretion signal of pro-UK was correctly processed.     

小麦品种抗性对麦长管蚜种群增长的影响

本研究采用室内、外结合的方法,分苗期和拔节期,对大田抗蚜性鉴定中表现不同的10个小麦品种的抗性对麦长管蚜种群增长的影响进行了研究。研究结果显示,品种抗性对麦长管蚜的存活率、发育进度、繁殖力有一致的抑制作用;品种间抗蚜性存在显著差异。同时通过组建不同品种上麦长管蚜繁殖特征生命表,从量的角度揭示了小麦品种抗性与麦长管蚜种群增长之间的关系。     

水平回转对水稻幼苗叶细胞的影响

对在模拟微重力装置上回转１４ 天的水稻幼苗叶细胞进行了亚显微形态、电子探针和细胞酶化学研究。发现叶细胞质膜上Ｃａ２＋ －ＡＴＰ酶活性消失,膜内钙总量上升、膜外钙总量下降,细胞骨架变得疏松,细胞壁变薄并凹凸不平。叶绿体的基粒和线粒体的内嵴亦有部分变化。其变化机制,首先是细胞质膜上Ｃａ２＋ －ＡＴＰ酶活性消失,膜上钙泵停止工作,跨膜钙浓度差减小,膜内钙浓度上升,微管、微丝聚合受阻,细胞骨架疏松,分泌泡移动失去导向,从而导致细胞壁变薄等状态

民勤红砂岗地区绵刺分布和繁殖方式及濒危原因调查

本文调查了民勤红砂岗地区绵刺的分布现状和面积,初步观测了它的生活习性和繁殖方式,发现枝条生根是它的重要繁殖方式,并对绵刺濒危的原因和保护措施进行了讨论。     

辐射增敏剂增敏活性的分子连接性研究

计算４２个辐射增敏剂的各阶分子连接性指数ｍＸｔ及△ｍＸｔ,并对其中３８个非对称性化合物的分子连接性指数与其增敏活性进行定量构效关系（ＱｕａｎｔｉｔａｔｉｒｅＳｔｒｕｃｔｕｒｅＡｃｔｉｖｉｔｙＲｅｌａｔｉｏｎｓｈｉｐ,ＱＳＡＲ）的研究,得到相关方程。并分析了影响增敏活性的１ｇＰ、ＥＳ及σ在一定程度上都可通过分子连接性指数表达出来。对这些增敏剂进行分子对称性的研究,发现对称性会降低分子的极性,进而降低其增敏活性。另外,还发现在这些增敏剂中存在亚类现象,并对各亚类的反应机理进行了探讨。
